18-54, SF is a rat cell line which grows under serum-free conditions and secretes rIGF-II. Concentrations of 75-100 ng/ml can accumulate in conditioned media. It was observed in this study that exogenous IGF-II increased cell numbers, whereas neither IGF-I nor insulin (both of which are structurally and functionally related to IGF-II) had this effect. Additionally, it was demonstrated that the cells possess classical IGF-II receptors, which are known to be identical with mannose-6-phosphate receptors. The cells also display specific receptors for both insulin and IGF-I. An anti-IGF-II receptor antibody inhibited cell proliferation. These results are consistent with an autocrine role for IGF-II which is mediated via its own receptor.;By screening the genomic library, the IGF-II gene was isolated. It contains a 5{dollar}\sp\prime{dollar} upstream region that has not previously been studied. Restriction mapping studies demonstrated that it does not differ from IGF-II genes identified in other rat cell types. The isolated gene should be useful for studying IGF-II gene regulation in these cells.
