The aim of this work is to provide a theoretical framework in which to relate the existing results of varying types of experiments at the subcellular level (patch clamp, vesicle studies, isotope tracer) with each other and with experiments performed at the cellular and epithelial levels (inhibitor response, electrical potential measurements, fluorescence microscopy) in order to derive a consistent and coherent view of the acid generating machinery of the stomach, with emphasis placed on the role of the acid secreting cell--the oxyntic cell. Partial differential equations describing the flow of electrolytes in the gastric microcirculation are derived; simplified subsets of these equations are solved analytically. Three areas have served as foci of investigation: (1) the mathematical representation of cellular ion fluxes and corresponding issues of stability, (2) a whole stomach model based on the balance between acid generation and self-protection and its correlations with experimental data, and (3) the way in which future experiments may or may not distinguish between different theoretical mechanisms of oxyntic cell acid secretion.
