Abstract

The main goals of this work are: to characterize the function of the mouse Hox-1.3 gene; to analyze its evolutionary conservation; and to provide a novel strategy for studying genes involved in the embryogenesis of Drosophila melanogaster. The developmental system of Drosophila melanogaster has several characteristics that make it an excellent experimental subject. This study reflects an effort to combine biochemistry, molecular biology, and classical genetics tools to achieve the goals. Accordingly, the specific aims of my research are: (1) to examine the evolutionarily conserved relationships between the mouse Hox-1.3 gene and its fly counterpart, Sex combs reduced; (2) to define the functional motifs of the mouse Hox-1.3 gene in cultured cells and in the transgenic flies system; (3) to study Hox-1.3 protein-DNA interactions in vitro; and (4) to utilize a targeted ribozyme as a potential tool for studying gene function in Drosophila.
