An efficient synthesis of an optically active inositol derivative that is a precursor to scD-myo-inositol 1,4,5-trisphosphate (scD-IP{dollar}\sb3{dollar}) is described. Regioselective crystallization of inositol diastereomers D4P and L4M from methanol followed by alkaline hydrolysis gives the two enantiomers ({dollar}-{dollar})-6-O-benzyl-2,3:4,5-di-O-isopropylidene-myo-inositol and (+)-4-O-benzyl-1,2:5,6-di-O-isopropylidene-myo-inositol. The ease with which these enantiomerically pure inositol derivatives were isolated facilitates the synthesis of more complex inositol phosphate derivatives such as scD-myo-inositol 1,3,4,5-tetrakisphosphate. The preparation of an activated inositol phosphate (3HP) and its use in the synthesis of a fluorescent analog of phosphatidylinositol and phosphatidylinositol 4,5-bisphosphate is also described.
